Amidites

Amidites are protected nucleotide derivatives containing a phosphoramidite functional group. Also known as phosphoramidites, they are highly reactive chemical intermediates widely used in the synthesis of oligonucleotides, DNA, RNA, and modified nucleic acid sequences. They play a central role in modern biotechnology, pharmaceutical development, molecular diagnostics, and genetic research. Due to their excellent coupling efficiency and compatibility with automated synthesizers, amidites have become the industry standard for solid-phase oligonucleotide synthesis. They enable the precise and stepwise assembly of nucleotide chains required for applications such as PCR primers, antisense oligonucleotides, siRNA, mRNA therapeutics, CRISPR guide RNA, and DNA sequencing technologies.

Types

  • DNA amidites. DNA amidites are essential building blocks used in the synthesis of standard DNA oligonucleotides. They contain protected nucleotide bases such as adenine, cytosine, guanine, and thymine, which enable accurate and efficient stepwise DNA chain assembly during solid-phase synthesis. Due to their high coupling efficiency and compatibility with automated synthesizers, DNA amidites are widely applied in PCR primer production, gene synthesis, DNA sequencing, molecular diagnostics, and biotechnology research.
  • RNA amidites. RNA amidites are specially designed nucleotide derivatives used for RNA oligonucleotide synthesis. Unlike DNA amidites, they include additional protection groups for the 2'-hydroxyl group of ribose sugars, ensuring stability during synthesis. RNA amidites are commonly used in the production of siRNA, mRNA therapeutics, CRISPR guide RNA, aptamers, and RNA interference research. Because of their moisture sensitivity and more complex chemistry, they require careful storage and controlled deprotection conditions.
  • Modified amidites. Modified amidites are functionalized phosphoramidite compounds developed to introduce specific chemical properties into oligonucleotides. These amidites can enhance molecular stability, improve binding affinity, or add functional groups for labeling and detection purposes. Common examples include fluorescent amidites, biotin amidites, amino modifiers, thiol modifiers, spacer amidites, and LNA amidites. They are extensively used in advanced diagnostics, therapeutic oligonucleotides, biosensors, fluorescence assays, and modern molecular biology applications.

Applications

  • Pharmaceutical industry. Amidites are essential raw materials in the development and manufacturing of modern oligonucleotide therapeutics. They are widely used in the synthesis of antisense oligonucleotides (ASOs), siRNA drugs, mRNA therapeutics, and gene-editing components.
