Poly(3-hydroxybutyrate-co-3-hydroxyvalerate), PHBV, is a microbial biopolymer with excellent biocompatibility and biodegradability. Due to the excellent physicochemical properties and relatively slow degradation rate, PHBV has been investigated as a platform for long-term drug release, for example, to encapsulate antineoplastic drugs for cancer therapy. Typical structures of polymeric micelles representing hydrophobic drug encapsulation and examples of polymers used as hydrophilic corona and hydrophobic core-PHBV are shown as figure.
This review focuses on the use of PHBV in tumor targeting with highlights including:
> Considering that drugs are usually entrapped in the amorphous regions of the polymeric material, PHBV copolymers are more suitable for higher drug loading than PHB homopolymers because the presence of hydroxyvalerate monomer in the polymer backbone can reduce crystallinity.
> PHBV nanoparticles are able to penetrate injured skin tissue and incorporate into cells as topical and transdermal drug delivery vehicles.
> Drug release kinetics from PHBV nanoparticles was related to polymer weight. Nanoparticles prepared with longer chain polymers generally exhibit slower drug diffusion and lower rates of hydrolytic degradation.
> The cytotoxic effect of PHBV nanoparticles was delayed compared to other polyester-based nanoparticles, which was associated with slower degradation of PHBV.
