α-Copaene exhibits potent antibacterial activity against major foodborne pathogens, including Staphylococcus aureus, Escherichia coli, Bacillus cereus, and Shigella bogdii. With a minimum inhibitory concentration (MIC) of 0.5-1 μL/mL and a minimum bactericidal concentration (MBC) of 2-4 μL/mL, α-copaene effectively disrupts bacterial cell membranes, leading to increased permeability and structural damage, as confirmed by scanning electron microscopy and flow cytometry. Moreover, α-copaene inhibits biofilm formation by reducing bacterial cell surface hydrophobicity, further preventing microbial adhesion and persistence. Hemolytic and cytotoxicity assessments indicate its biosafety, supporting its potential as a food-grade preservative. In practical application, α-copaene at 4 μL/mL effectively inhibited bacterial growth in beef soup, demonstrating its efficacy in real food matrices.
