2,4-di-tert-butylphenol (2,4-DTBP) is a natural product with diverse physiological activities and regulatory effects on the soil environment. In this study, we investigated its potential to control bacterial wilt caused by the plant pathogen Ralstonia solanacearum, and the molecular and functional mechanisms involved.
Mechanism of Action
2,4-DTBP significantly restricts bacterial proliferation and induces morphological alterations. The compound impedes key virulence mechanisms including biofilm formation, motility, and exopolysaccharide (EPS) secretion. Transcriptomic analyses indicate inhibition of energy production and membrane transport systems. 2,4-DTBP increases membrane permeability, induces reactive oxygen species (ROS) accumulation, and promotes electrolyte leakage, leading to loss of cellular viability.
Key Findings
The results showed that 2,4-DTBP treatment of infected plant roots significantly reduced bacterial biomass, indicating that the pathogenicity of the pathogen was effectively reduced. The bacterial membrane was damaged and some important metabolic pathways were inhibited. 2,4-DTBP was also shown to have a multi-target inhibition effect, and it reduced the ability of the bacteria to infect and colonize the host plant.

2,4-Di-tert-butylphenol (2,4-DTBP) has emerged as a promising eco-friendly antifungal compound. This study explored the antifungal activity and mechanism of 2,4-DTBP against Ustilaginoidea virens, the causal pathogen of rice false smut, for potential application in sustainable crop protection.
Mechanism of Action
2,4-DTBP exerts its antifungal effect by causing damage to the fungal cell wall and membrane. It disrupts cellular redox homeostasis and leads to cell death. Transcriptomic analysis identified 353 upregulated and 367 downregulated genes. Processes like spindle microtubule assembly, cell wall and membrane integrity, cellular redox homeostasis, mycotoxin biosynthesis, and intracellular metabolic processes were significantly affected.
Highlights
The compound inhibited the growth of U. virens at a low concentration (EC50 = 0.087 mmol/L). Observation by microscopy and biochemical assays indicated structural damage and loss of cell viability. It suppressed virulence factors such as mycotoxin production.
