The adsorption and immobilization of lipase on silica nanoparticles were investigated to enhance enzyme stability and activity. In the isothermal adsorption experiment, varying concentrations of lipase were added to silica nanoparticles in a controlled water bath environment, allowing adsorption to occur at 25°C with constant stirring. The equilibrium adsorption was assessed by measuring the lipase concentration in the solution post-centrifugation. The results revealed a clear adsorption curve, indicating the effectiveness of silica nanoparticles in adsorbing lipase, with the amount of adsorbed enzyme increasing with concentration.
Further, in the immobilization process, lipase solutions were prepared and mixed with silica nanoparticles in a buffer solution, followed by shaking at 35°C for three hours. The process significantly increased the immobilization efficiency, with an optimal recovery rate of hydrolytic enzyme activity observed at specific nanoparticle dosages.
