Glucan endo-1,3-β-D-glucosidase (EC 3.2.1.39), encoded by the PcENDO1 gene, functions as a core cell-wall-remodeling hydrolase in Phytophthora cinnamomi, supporting hyphal growth, cell wall metabolism, and infection progression in chestnut ink disease of Castanea sativa.
Assay Protocol: Genomic DNA was isolated from P. cinnamomi mycelia. A 1231 bp fragment was amplified by standard PCR using conserved primers. The full 2586 bp PcENDO1 gene was obtained via thermal asymmetric interlaced (TAIL)-PCR with nested gene-specific primers and degenerate primer R3. Sequence characterization included BLAST alignment, conserved domain analysis, and phylogenetic comparison.
Performance Evaluation: PcENDO1 belongs to glycoside hydrolase family 17 with a canonical catalytic domain. It shows high homology to Phytophthora infestans endo-1,3-β-glucanase (E-value = 8e-135). A conserved 16-bp oomycete transcriptional motif was identified 68-83 nt upstream of the ATG start codon. This enzyme is essential for cell wall dynamics and host-pathogen interaction in destructive ink disease.
