Nonivamide, a TRPV1 agonist, was investigated for its effects on human corneal epithelial cells (HCE-T). Subtoxic concentrations (100-200 μM) were applied, and cell proliferation was monitored using cell viability assays and impedance measurements. Flow cytometry revealed cell cycle arrest, while western blotting quantified p21 upregulation and cyclin D1 downregulation. Real-time fluorescence imaging assessed Ca²⁺ influx and mitochondrial membrane potential. Nonivamide induced oxidative stress through TRPV1-mediated Ca²⁺ influx, decreasing mitochondrial membrane potential and impairing proliferation. This study demonstrates a direct experimental application of nonivamide in evaluating corneal cytotoxicity and elucidates mechanistic pathways underlying ocular injury.

Nonivamide was applied to porcine subcutaneous pre-adipocytes at 25 μM to evaluate effects on adipogenesis and browning. Post-differentiation, western blotting and gene expression analyses quantified Pparγ, Cebpα, Hsl, Atgl, and brown/beige adipocyte markers (Prdm16, Cidea, Slc27a1). Thermogenesis-related genes Cpt1a and Cpt1b, and mitochondrial biogenesis markers Tfam, Nrf1, Nrf2, and Tomm20 were assessed. Nonivamide inhibited adipogenesis, promoted lipolysis, and induced browning, demonstrating its utility as a tool for experimental modulation of adipocyte phenotype and mitochondrial activity in obesity research.

Nonivamide was incorporated into castor oil-based dispersed phases of oil-in-oil emulsions containing polydimethylsiloxanes and silicone surfactants. The influence of nonivamide content, phase volume ratio, and silicone oil viscosity on in vitro release and skin permeation was measured. Permeation studies demonstrated that formulation variables controlled nonivamide release, achieving stable release over 10 h. This experimental approach highlights nonivamide's application as an active pharmaceutical ingredient in emulsion systems for sustained dermal delivery, providing a model for optimizing controlled-release formulations.

Nonivamide (50 μM) was combined with hyperthermia (44 °C, 15 min) to assess apoptosis in U937 cells. Flow cytometry, ROS measurement, mitochondrial potential assays, and western blotting were employed to evaluate apoptotic pathways. Results showed enhanced ROS generation, mitochondrial dysfunction, cleaved caspase-3 activation, increased pro-apoptotic Bcl-2 family proteins, decreased anti-apoptotic Bcl-2, and upregulation of p-JNK and p-p38. This study demonstrates the experimental use of nonivamide as a non-toxic adjuvant to potentiate hyperthermia-induced apoptosis via mitochondrial-caspase and MAPK signaling pathways in cancer research.
