Thymidine-derived functional monomers demonstrated clear experimental value in constructing molecularly imprinted polymers (nanoMIPs) for adenosine sensing. In this study, acrylamide-modified and carboxy-modified thymidine monomers were incorporated into nanoMIP synthesis via solid-phase imprinting. Two immobilization strategies-glutaraldehyde linking and EDC/NHS coupling-were compared to evaluate their impact on template orientation and binding performance. Surface plasmon resonance (SPR) was employed as the primary analytical method to quantify binding affinities, revealing that thymidine-based nanoMIPs produced via the glutaraldehyde method achieved the highest affinities, with KD values as low as 2.11 nM. Selectivity experiments against structurally related nucleosides confirmed precise molecular recognition. Furthermore, the nanoMIPs maintained robust binding behavior in fetal bovine serum, demonstrating the practical applicability of thymidine-modified monomers in developing rapid, high-specificity biosensing platforms.
