Sensitive and rapid analysis of DNA fragments, particularly for pathogen detection, is essential in biomedical and food safety applications. In this study, tetrabutylammonium phosphate (TBAP) was employed as an additive in a non-gel sieving capillary electrophoresis (NGS-CE) method using hydroxypropylmethylcellulose (HPMC) as the sieving matrix. The tetrabutylammonium cation interacts electrostatically with negatively charged DNA, improving fragment resolution without interfering with UV detection at 260 nm.
Key Results:
· The addition of 15 mM TBAP significantly improved resolution for all DNA fragments (100-1000 bp), with resolution values increasing by 20-40% compared to TBAP-free conditions. Field-enhanced sample injection (FESI) provided 25- to 52-fold sensitivity enhancement, achieving detection limits as low as 2.2-5.8 ng/mL.
· The method successfully separated three multiplex PCR products (from L. monocytogenes, E. coli, and S. enterica) and their corresponding primers in 25 minutes with baseline resolution.
· High reproducibility was demonstrated, with intraday RSDs for migration time and peak area below 2.4% and interday RSDs below 6.1%. PCR product sizes could be accurately estimated using a migration time-based calibration curve, with deviations ≤1.8%.
· TBAP thus serves as an effective, non-absorbing ion-pair reagent that enhances both resolution and sensitivity in DNA analysis by CE-UV, offering a practical tool for diagnostic and food safety applications.
