The remediation of sites co-contaminated with chlorinated solvents (e.g., TCE) and aromatic hydrocarbons (e.g., BTEX) is often complicated by the presence of Per- and Polyfluoroalkyl Substances (PFAS) from aqueous film-forming foams (AFFFs). Effective bioremediation relies on specific microbial communities, whose activity may be inhibited by PFAS. This study investigated the inhibitory potential of the PFASs including perfluorohexanesulfonamide (FHxSA) on key microorganisms involved in anaerobic and aerobic biodegradation processes.
Testing Method: The impact of FHxSA was evaluated in parallel with other surfactants, including its zwitterionic precursor (AmPr-FHxSA), the anionic perfluorohexanesulfonic acid (PFHxS), and a non-fluorinated control (SDS). Individual PFAS were applied at concentrations of 1-10 μM. Three distinct microbial cultures were exposed.
Key Findings:
· Both FHxSA and its precursor AmPr-FHxSA inhibited the aerobic BTEX-degrading culture. This inhibitory effect was more pronounced than that caused by the anionic PFHxS at equivalent concentrations. The anaerobic TCE-dechlorinating coculture was not inhibited by individual PFAS exposures, including FHxSA. Similarly, the anaerobic toluene-degrading culture showed no inhibition from individual PFASs or diluted AFFF.
· Metabolite analysis indicated that exposure to surfactants, including FHxSA, altered amino acid concentrations in the cultures. The aerobic BTEX culture showed lower concentrations of all amino acids upon exposure, while the anaerobic TCE culture exhibited an increase, suggesting different stress response mechanisms potentially related to cell membrane synthesis and protein stress signaling.
