N-Acetylimidazole in the Functional Modulation of Multicatalytic Proteinase Complex: A Casein Hydrolysis Application

Chemical modification of the bovine pituitary multicatalytic proteinase complex by N-acetylimidazole. Reversible activation of casein hydrolysis. Yu B, et al. Journal of Biological Chemistry, 1991, 266(26), 17396-17400.

N-Acetylimidazole was experimentally applied to modulate the catalytic profile of the bovine pituitary multicatalytic proteinase complex (MPC). The study employed substrate-specific fluorometric assays to monitor three proteolytic activities: trypsin-like (Cbz-D-Ala-Leu-Arg-2-naphthylamide), peptidylglutamyl-peptide hydrolyzing (PGP; Cbz-Leu-Leu-Glu-2-naphthylamide), and chymotrypsin-like (Cbz-Gly-Gly-Leu-p-nitroanilide). Upon acetylation with N-acetylimidazole, a rapid inactivation of the trypsin-like and PGP activities was observed, while chymotrypsin-like activity declined slowly. Paradoxically, casein hydrolysis was significantly enhanced, yielding a stable 21 kDa intermediate not present in native reactions. Subsequent treatment with hydroxylamine restored the trypsin-like and caseinolytic activities but not PGP, confirming the irreversible inhibition of glutamyl-specific catalysis via ε-lysine acetylation. Non-denaturing gel electrophoresis revealed mobility shifts without subunit dissociation, consistent with lysine-targeted acetylation. These results demonstrate that N-acetylimidazole can differentially modulate MPC's enzymatic specificity and suggests a distinct protease component is responsible for initiating casein cleavage, separate from classical proteolytic activities.
