L-Formiminoglutamic Acid as a Specific Substrate for Trypanosoma cruzi Formiminoglutamase

Pathways I-III for L-histidine catabolism containing L-formiminoglutamic acid as key intermediate. Hai, Yang, et al. Biochemistry 52.51 (2013): 9294-9309.

L-Formiminoglutamic acid is the key intermediate in histidine utilization pathway I. It serves as the natural substrate for formiminoglutamase (FIGase), which hydrolyses its formimino group to yield L-glutamate and formamide. This reaction completes the catabolism of L-histidine to glutamate in many bacteria and protozoa, including Trypanosoma cruzi.
Enzymatic Assay: The activity of TcFIGase was measured colorimetrically at pH 9.5 using L-formiminoglutamic acid as the substrate. The reaction was stopped with saturated sodium borate solution, followed by addition of a chromophore reagent (sodium nitroprusside, potassium ferricyanide and NaOH). Absorbance was read at 485 nm. Initial velocities were fitted to the Michaelis-Menten equation.
Performance Evaluation: Wild-type TcFIGase (expressed in LB media) exhibited a kcat of 200 ± 20 s-1, a KM of 40 ± 10 mM and a catalytic efficiency kcat/KM of 5200 ± 200 M-1 s-1. The enzyme showed no activity towards other guanidino substrates such as L-arginine, agmatine or guanidinobutyric acid, confirming its strict specificity for L-formiminoglutamic acid. Reconstitution of the apoenzyme with Mn2+ restored maximal activity, whereas other divalent metals (Co2+, Ni2+, Mg2+) gave lower activities.
L-Formiminoglutamic acid is a specific, high-KM substrate for TcFIGase. Its hydrolysis depends on a binuclear manganese cluster and a positively charged arginine residue (R144) that likely binds the side-chain carboxylate. This substrate-enzyme pair is a valuable model for studying the structure and mechanism of the ureohydrolase superfamily.
