In this study, Gallium(III) acetate was evaluated for its antimicrobial potential against antibiotic-resistant Pseudomonas aeruginosa. Using an integrative approach combining metalloproteomics, metabolomics, and transcriptomics, RNA polymerase subunits RpoB and RpoC were identified as direct Ga(III)-binding targets. Functional assays demonstrated that Ga(III) acetate suppresses RNA synthesis, decreasing bacterial metabolic activity and energy utilization. Experimental co-treatment with exogenous acetate further potentiated antimicrobial efficacy, enhancing Ga(III) uptake and disrupting TCA cycle flux and bacterial respiration. Persister cell growth and virulence were significantly reduced in both mammalian cell assays and murine skin infection models. These results highlight Gallium(III) acetate as a mechanistically validated antimicrobial agent and demonstrate a practical metabolite-based strategy to enhance Ga(III) activity in combating drug-resistant bacterial infections.
