Gadolinium-based contrast agents are widely used in magnetic resonance imaging (MRI), but conventional molecular chelates suffer from relatively low relaxivity and potential toxicity concerns. In this study, 3,5-diiodo-L-tyrosine (DLT) was employed as a bifunctional coating ligand for ultrasmall Gd2O3 nanoparticles to develop a potential MRI contrast agent with improved relaxivity and biocompatibility.
DLT-coated Gd2O3 nanoparticles (DLT-NPs) were synthesized via a one-pot polyol method in triethylene glycol, followed by surface modification with DLT. DLT serves as an effective bifunctional coating ligand for ultrasmall Gd2O3 nanoparticles, providing colloidal stability, high r1 relaxivity, and biocompatibility for potential MRI contrast agent applications.
Key Results:
· HR-TEM revealed ultrasmall, spherical DLT-NPs with an average diameter of 2 nm, suitable for potential renal clearance (<3 nm hydrodynamic diameter). FT-IR confirmed successful surface coating.
· DLT-NPs exhibited superior longitudinal relaxivity: r1= 9.24 /s/mM at 1.5 T, significantly higher than commercial Gd-DTPA (r1 ≈ 3.5-4.0 /s/mM). Concentration-dependent brightening in T1-weighted MR images confirmed positive contrast enhancement.
· The nanoparticles remained paramagnetic down to 5 K, with saturation magnetization of 177.78 emu/g at 5 K and 6.63 emu/g at 300 K.
· Cytotoxicity assays showed >60% cell viability for both DU145 and NCTC1469 cell lines at Gd concentrations up to 50 μM, with negligible toxicity below 10 μM, indicating good biocompatibility.
