Vasarenes - vase-shaped cage compounds derived from ninhydrin + phloroglucinol - are known to assemble into supramolecular cages that encapsulate alkali fluoride ion-pairs. To examine how framework geometry affects ion-pair recognition, the original three-walled vasarene motif was systematically modified by replacing the phloroglucinol unit with other polyhydroxy aromatics. 1,2,4,5-benzenetetrol was selected as one such alternative building block to produce "two-walled" vasarene analogues that retain large, voluminous cavities while altering spatial arrangement of hydrogen-bond donors and hemiketal formation sites.
Synthesis Details: Ninhydrin (565 mg, 3.17 mmol) and 1,2,4,5-benzenetetrol (250 mg, 1.76 mmol) were stirred in glacial acetic acid (35 mL) at 80-90 °C for 24 h. Cooling to room temperature, a yellowish precipitate was formed. The solid was collected by filtration, washed with glacial acetic acid and dried in vacuo to give the bis-ninhydrin adduct as a yellow powder (560 mg, 75% yield). Crystals were obtained by crystallization from methanol as colorless crystals.
