3-(3-Triethoxysilylpropyl)oxolane-2,5-dione serves as a key silanization reagent for surface functionalization of tapered singlemode-no core-singlemode (SNS) fiber biosensors, enabling stable covalent immobilization of antibodies for high-sensitivity Staphylococcus aureus detection.
Modification Protocol: The tapered SNS fiber (taper waist diameter 10.2 μm) was cleaned with ethanolic KOH solution, then silanized with 3-(3-triethoxysilylpropyl)oxolane-2,5-dione for 4 hours. Its anhydride group hydrolyzed spontaneously to carboxyl groups, followed by activation with EDC/NHSS. Pig IgG antibodies were immobilized, and BSA was used to block non-specific binding sites.
Performance Evaluation: This silane reagent efficiently introduced reactive carboxyl groups onto the fiber surface, ensuring stable antibody conjugation and reducing non-specific biofouling. The functionalized biosensor reached a refractive index sensitivity of 1127.3 nm/RIU and achieved a 1.1 nm wavelength blue shift toward S. aureus at 101 CFU/mL, realizing ultra-sensitive detection of foodborne pathogens.
