Evaluating the Role of Impurities in Disperse Blue 124 Contact Allergy Diagnosis

Disperse Blue (DB) 106 and DB 124 are recognized as significant contact sensitizers among textile dyes, frequently identified among the most common disperse dye allergens. Due to their allergenic potential and structural similarity (leading to frequent co-reactivity), they are often recommended as screening allergens for textile dye dermatitis. This investigation aimed to determine the significance of impurities present in commercial DB 106 and DB 124 patch test preparations concerning the diagnosis of contact allergy to these dyes.
Twenty-one patients diagnosed with contact allergy to DB 106 and/or DB 124 underwent patch testing with dilution series of commercially available and purified samples of DB 106 and DB 124.
Key Findings
· TLC Strip Reactivity: Positive reactions occurred to TLC strips of commercial DB 106 in 13 out of 21 patients and to DB 124 strips in 12 out of 18 patients tested.
· Reactivity to Impurities: Among the patients reacting to the TLC strips, 4 out of 13 (DB 106) and 5 out of 12 (DB 124) reacted only to spots corresponding to impurities, not to the main spot representing the dye itself.
· Purified vs. Commercial Dye Reactivity: Dilution series testing revealed that purified DB 106 elicited positive reactions in 10 patients, while the commercial DB 106 preparation elicited reactions in 16 patients. Similarly, purified DB 124 elicited reactions in 10 patients, whereas the commercial DB 124 preparation elicited reactions in 15 patients.
