Bulk powders of sulfamethazine (SMZ) polymorph II with narrow particle size distribution were prepared and two known sulfamethazine polymorphs I and II were physically characterized. Sulfamethazine sodium salt (NaSMZ) was used to achieve rapid crystallization of SMZ from water. The results showed that the two polymorphs were enantiomers with slow kinetics of interconversion. The thermodynamic transition temperatures were between 51 and 54°C, with polymorph II being stable at lower temperatures.
NaSMZ used in the preparation of polymorph II
· SMZ polymorph II can be obtained by recrystallization from a solution prepared by suspending an excess of polymorph I in acetonitrile. In addition, when the corresponding seed crystals are available, a large amount of polymorph II can be more efficiently crystallized from acetonitrile by a slurry process.
· During the recrystallization process, rapid crystallization of SMZ from water was achieved by adding hydrochloric acid to an aqueous solution of SMZ sodium salt. The crystals thus obtained were washed thoroughly with distilled water to remove residual electrolytes. This method is called "sodium salt acidification".
