Assay Protocol: Neutral ceramidase hydrolyzes NBD-C12-ceramide to release NBD-dodecanoic acid. Glomerular lysates (50 μg protein) are incubated with 25 μM NBD-C12-ceramide at 37℃ for 2 h. Lipids are extracted with chloroform/methanol (1:1), separated via TLC, and NBD-dodecanoic acid is quantified by fluorescence imaging.
Performance Evaluation: This substrate supports specific, quantitative N-CDase activity measurement (pmol/min/mg protein). In STZ-induced diabetic rats, it accurately detects elevated glomerular N-CDase activity, which correlates with S1P accumulation and early glomerular mesangial cell proliferation. It avoids non-specific interference, facilitating mechanistic studies of sphingolipid metabolism in early diabetic renal injury.

Assay Protocol: Recombinant zebrafish AC expressed in Pichia pastoris GS115 was incubated with C12-NBD-ceramide substrate in assay buffer (pH 5.6) at 37℃ for 90 min. Lipids were extracted with chloroform/methanol, separated on silica gel 60 TLC plates, and observed under UV light. NBD-dodecanoic acid was used as the positive control to confirm product formation.
Performance Evaluation: This fluorescent fatty acid standard presents a stable Rf value of 0.71 on TLC, distinctly separated from the C12-NBD-ceramide substrate (Rf=0.92).
