Anti-Leishmania Activity of Lipophilic N-Alkyldiamines Such as N-Decyl-1,3-Propanediamine

Researchers synthesized a group of long-chain N-monoalkylated diamines (like N-decyl-1,3-propanediamine) and two pyridinediamine derivatives and tested their anti-Leishmania effectiveness in vitro.
Key Findings
The ethylenediamine derivative 10 (N-dodecyl-1,2-ethanediamine), featuring a 12-carbon alkyl chain, demonstrated the highest antiproliferative efficacy against both L. amazonensis and L. chagasi, with IC50 values of 0.94 μM and 0.26 μM, respectively. Notably, it exhibited 7.3-fold greater activity against L. chagasi compared to the reference drug amphotericin B. Within the same series, compounds 8, 9, and 11 also displayed strong in vitro activity against L. amazonensis promastigotes (IC50 < 8 μM). Every ethylenediamine derivative except for compound 7 demonstrated effectiveness against L. chagasi promastigotes with IC50 values under 5 μM.
Among N-alkylated propanediamine derivatives, compounds 15 (N-decyl-1,3-propanediamine), 16, and 17 exhibited activity against both parasite forms, with 15 being the most potent (2.6 times more active than amphotericin B against L. chagasi; IC50 = 3.0 μM for 14). Butanediamine and hexanediamine derivatives (20 [N-dodecyl-1,4-butanediamine] and 22) displayed notable activity against L. amazonensis and exceptional efficacy against L. chagasi. Conversely, N-methyl-pyridyl derivatives 23 and 24 showed no activity at tested concentrations.
