α-Casein(90-95) is a bioactive peptide derived from bovine α-casein. It acts as a non-IgE-dependent mast cell activator, inducing degranulation and release of inflammatory mediators such as histamine and tryptase. This property makes it a useful reference for assessing the allergenicity of food-derived peptides. α-Casein(90-95) is a potent, non-IgE-mediated mast cell activator.
Experimental Protocol: Mast cells were obtained from the peritoneal cavity of rats or differentiated from mouse bone marrow using conditioned medium. Cells were incubated with α-casein(90-95) at milli- to micromolar concentrations. Degranulation was quantified by measuring histamine release (ELISA) and tryptase activity (chromogenic substrate tosyl-Gly-Pro-Lys-p-nitroanilide). Calcium ionophore A23187 served as a positive control.
Performance Evaluation: α-Casein(90-95) elicited a significant secretagogue effect, causing marked histamine and tryptase release from peritoneal mast cells at micromolar levels. In contrast, β-casomorphin-7 required higher concentrations to achieve comparable effects on bone-marrow-derived mast cells. The response was independent of IgE, resembling that of anaphylatoxins. No detectable degranulation was observed in negative controls.
