Silicon 2,3-naphthalocyanine bis(trihexylsilyloxide) (SiNc) is a high-performance functional dye with significant potential in ophthalmic applications. SiNc was integrated into poly(HEMA-co-GMA)-based copolymer hydrogel systems via thermal polymerization. Its unique absorption characteristics enable selective blocking of UV-A and UV-B radiation while maintaining high visible light transmittance and material compatibility.
Material Preparation and Polymerization
The monomer system used for the preparation of the specimens included 2-hydroxyethyl methacrylate (HEMA), MMA, n-vinyl pyrrolidone (NVP), ethylene glycol dimethacrylate (EGDMA) and with different contents of glycerol dimethacrylate(GMA, 1~10%). The concentration of SiNc was 1 wt% and the preparation process of it was uniformly dispersed by ultrasonic treatment. The polymerization initiator was AIBN and the polymerization method was cast mould. The hydration of the samples was in the 0.9% saline solution for 24 hours before the experiments were carried out.
Key Performance Results
The visible light transmittance of G_SN samples (SiNc in Ref._SN) ranged from 86.0% to 89.4%, UV-B from 63.0% to 67.6%, and UV-A from 62.5% to 66.0%. The spectral transmittance measurement showed that SiNc only absorbed a small amount of UV light.
It can be concluded that incorporating SiNc into ophthalmic polymer materials could be beneficial for producing colored ophthalmic lenses that provide protection against UV and infrared light while maintaining their physical properties.

This study evaluated its performance as a highly efficient contrast agent and functional reporter in both in vitro and in vivo settings, emphasizing its advantages over conventional agents such as indocyanine green (ICG).
Preparation of Naphthalocyanine Solution
SiNc was prepared as an emulsion of 10% Cremophor EL, 1% 1,2-propanediol, and 1% dimethylformamide in phosphate buffered saline pH 7.4 with sonication. The concentration of the starting solution was calculated based on a measurement of the absorbance of the solution with a molar extinction coefficient of 500,000 M-1 cm-1 resulting in an approximate concentration of 40 µM.
Methods & Results
· In Vitro Analysis: Photostability, singlet oxygen yield, and optoacoustic response were measured under pulsed laser irradiation. SiNc generated significantly stronger optoacoustic signals than indocyanine green. Minimal photodegradation was observed under repeated laser exposure.
· In Vivo Evaluation: HT-29 tumor-bearing mouse models were used to assess biodistribution and contrast enhancement via multispectral optoacoustic tomography (MSOT). MSOT clearly resolved SiNc accumulation within tumors and correlated with perfusion parameters.
· The compound demonstrated singlet oxygen generation under laser illumination, supporting its use in photodynamic therapy and treatment monitoring.
