2,3,4,5,6-Pentafluorobenzoic acid was employed as a key functional reagent for constructing embedded polar group (EPG)-containing pentafluorophenyl (PFP) stationary phases on silica, aiming to enhance chromatographic selectivity beyond conventional C18 materials. The silica surface was first functionalized with (3-aminopropyl)trimethoxysilane (APS) to introduce reactive amino groups. Subsequently, APS-modified silica (3.0 g) was reacted with 2,3,4,5,6-pentafluorobenzoic acid (3.00 g, 0.014 mol) in methanol (50 mL) in the presence of diethyl pyrocarbonate (DEPC, 1.5 g, 0.009 mol) and triethylamine (TEA, 1.1 g, 0.011 mol) at 60 °C for 24 h to form the Sil-PFP-BA stationary phase. The product was purified by repeated methanol centrifugation and vacuum drying. Experimentally, 2,3,4,5,6-pentafluorobenzoic acid enabled stable covalent immobilization of the PFP moiety, providing additional π-π, dipole-dipole, and polar interaction sites that significantly improved chromatographic separation performance for structurally similar analytes.
