Sulfur (VI) Fluorides
Sulfur (VI) fluorides are a class of click chemistry reagents containing S-F chemical bonds, which result in their unique properties such as hydrolytic stability and resistance to reduction. Initially, as covalent protein modifiers, they were applied to proteomics, protease inhibitors, and chemical probes. Until 2014, sulfur-fluoride exchange (SuFEx) click reaction were proposed by Sharpless and coworkers, the application of sulfur (VI) fluorides really took off. In contrast to other sulfur(VI) halides, sulfur(VI) fluorides react exclusively at the sulfur atom in the presence of nucleophiles primarily through a nucleophilic addition/elimination mechanism. Common sulfur (VI) fluorides include sulfonyl fluorides (RSO2F), fluorosulfates (ROSO2F), and sulfamoyl fluorides (R2NSO2F), which have important applications in chemical biology to polymers.
